Title: Connectome-based Spreading of Grey Matter Abnormalities Across the Psychosis Continuum.

Abstract: Decades of research and thousands of articles have established that psychotic disorders are associated with a complex array of grey matter reductions that can worsen with illness progression [1-3]. However, the mechanisms explaining the specific anatomical distribution of these changes have remained elusive. Different regions of the brain's grey matter are connected by a complex structural network of white-matter fibres, which are responsible for the propagation of action potentials and the transport of trophic and other molecules. In neurodegenerative disease, these connections constrain the way in which grey-matter pathology progresses [4-5]. Here we investigated whether connectome architecture also shapes the spatial pattern of cross-sectional and longitudinal grey matter volume changes across the psychotic illness continuum, using multiple independent datasets.
